You can take a train from Tokyo to Karuizawa-machi in 1.5 hours. Other options are to drive; or take a bus. Prepare for a bit cooler weather when traveling from Tokyo in October: high temperatures in Karuizawa-machi hover around 21°C and lows are around 14°C. Once a small fishing town, Yokohama was one of the first ports to welcome foreign trade in the 19th century, after which it grew into Japan's second largest city and a bustling metropolis.
Get out of town with these interesting Yokohama side-trips: Sojiji (in Nishitokyo), Kōtoku-in (in Kamakura) and Ghibli Museum Mitaka (in Mitaka). There's lots more to do: take in the spiritual surroundings of Hase-dera Temple, get up close and personal with brilliant marine life at Yokohama Hakkeijima Sea Paradise, contemplate the long history of Tsurugaoka Hachimangu Shrine, and get to know the fascinating history of Sankeien Gardens.

Do a combination of bus and train from Hakone-machi to Yokohama in 1.5 hours. Alternatively, you can drive; or take a bus. While traveling from Hakone-machi, expect somewhat warmer days and about the same nights in Yokohama, ranging from highs of 25°C to lows of 19°C. Wrap up your sightseeing by early afternoon on the 3rd (Sun) to allow time for travel back home.
